Differences in synthesis and degradation of serum proteins in normal and hepatoma-bearing animals.

چکیده
Rats bearing Morris liepatoma 7800 and Morris hepatoma 7777 have serum protein compositions which differ from normal rats. There is a slight decrease of albumin and an increase in a-globulin proteins in the circulating blood of the tumor-bearing animals. The increase in a-globulin is larger than would be expected if globulin would just compensate for the slight decrease in albumin, resulting in increased protein levels in the hepatoma-bearing animals. Synthesis of serum proteins released into the circulating blood was similar in host and normal rats. a-Globulin proteins in tumor-bearing rats had a decreased rate of breakdown, accounting for the absolute increase of these proteins. The capacity of liver tissue and hepatoma tissue to synthesize albumin differs 11-fold, with the hepatomas showing the lower rate of synthesis. In vitro determinations showed a 6to 7-fold difference. Host liver had the same synthetic capacity as normal liver. Both total protein synthesis and albumin synthesis are inhibited by cycloheximide and puromycin. The albumin synthesized by the hepatomas is apparently not released into the circulating blood to any great extent.

متن کاملHEPATOMA McARDLE-RH7777 CELLS HAVE THE SAME RESPONSE AS NORMAL RAT HEPATOCYTES TO BOTH DIBUTYRYL-cAMP AND ANTICALMODULINW-7
The effects of cAMP-analogue dibutyryl-cAMP and anticalmodulin W-7 were studied on de novo synthesis and secretion of lipids in cultures of hepatoma McArdle RH7777 cells and normal rat hepatocytes. Dibutyryl-cAMP and W -7 separately caused a significant decrease in the secretion of de novo synthesized triacyl [3H]glycerol in both cultures of McArdle cells and rat hepatocytes. The inhibito...
متن کاملIntracellular synthesis of alpha-fetoprotein and fibrinogen without secretion by Zajdela rat ascites hepatoma cells.
Transplabtable Zajdela rat ascites hepatoma cells, previously considered "nonproducers," synthesize detectable amounts of intracellular alpha-fetoprotein (AFP) and fibrinogen, but fail to secret or release these serum proteins. Evidence for defective secretory mechanisms for serum proteins in these hepatoma cells (a) explains the failure to detect AFP in either the serum or ascitic fluid of rat...

Background and purpose : The levels of serum acute phase reactant proteins change in different malignant diseases that along with other proteins could be assessed by electrophoresis. Materials and methods: The electrophoretic pattern of serum proteins of 85 patients bearing different types of neoplasia and 85 matched normal adults were comparatively studied by agarose gel electrophoresis t...
متن کاملKinetics of the acute-phase reaction in rats after tumor transplantation.
Transplantation of Yoshida sarcoma (solid type) and Zajdela ascites hepatoma tumors in rats induces a biphasic change in the concentration of the following five acute-phase proteins: alpha-1-acid glycoprotein; alpha-1-antitrypsin; haptoglobin; hemopexin; and ceruloplasmin. These proteins and other plasma proteins were quantitated by two-dimensional immunoelectrophoresis relative to normal serum...
